BACKGROUND:

In order to improve antioxidant and anti-fatigue capacities of the whey protein for wide utilization, hydrolysis was conducted by chymotrypsin (EC. 3.4.21.1). Different molecular weight (MW) range of whey protein hydrolysates (WPH) were separated by ultrafiltration (UF). Kunming mice in the treatment groups were orally administered whey protein isolate (WPI) or WPH (1.5 g kg-1 body weight) with different MW (<5 kDa; 5-10 kDa; 10-30 kDa; >30 kDa) for six weeks to explore whether different MW part in WPH affected mice fatigue.

RESULTS:

Compared with control group (orally administered 9 g kg-1 saline) or WPI group, WPH groups with low MW (<10 kDa) could prolong the swimming time (P<0.05), and demonstrated higher concentrations (P<0.05) of glucose, non-esterfied fatty acid (NEFA), liver glycogen, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and lower concentration of lactate. WPH with low MW (<10 kDa) had higher hydroxyl- and α,α-diphenyl-β-picrylhydrazyl (DPPH)-scavenging ability and Ferrous-chelating capacity compared with WPI.

CONCLUSION:

It proved that low MW WPH (<10 kDa) with higher anti-fatigue capacity demonstrated higher free radical scavenging capacity and ferrous chelating ability.
